Basic Facts About  Pyrimethamine
Fact 1: Pyrimethamine is used to treat toxoplasmosis and Pneumocystis pneumonia in HIV/AIDS patients. 

Fact 2: It is taken alongside leucovorin (folinic acid) due to its mode of effect (see below). 

Fact 3: It is no longer widely used to treat malaria. 

Fact 4: It is taken orally and is classed as pregnancy category C.

Fact 5: Side effects include: 
Nausea/Vomiting
Glossitis
Diarrhea
Skin rash
Thrombocytopenia
Bone marrow suppression
Ataxia
Tremors
Loss of appetite

Most of the above side effects manifest at high doses. 

Fact 6: Other antifolate drugs such as methotrexate and trimethoprim can increase the risk of folate deficiency and blood dyscrasias. Taking
pyrimethamine with folinic acid reduces these risks.

Fact 7: Mechanism of action -
#Pyrimethamine works by competitively inhibiting the enzyme, dihydrofolate reductase interfering in the generation of tetrahydrofolate (THF).
# THF is essential for DNA and RNA synthesis in many parasite organisms.
